Effect of oxytocin on reparative regeneration of corneal structures

The influence of Oxytocin in corneal mechanical damage was estimated, its role in medical correction from the perspective of reparative morphogenesis was determined. All experimental animals (6 rabbits) were divided into 3 groups. Corneal erosion, 8 mm in diameter, was created in the first and the second groups. The third group was control, without any damages of the cornea. The 1st group (2 rabbits) was treated by intraconjunctival instillation of antibiotic Cipromed, 0.3%; the 2nd group (2 rabbits) ― by Oxytocin 5 ME and Cipromed, 0.3%. Samples (n=18) of the anterior epithelium and stroma were taken from all animals at days 4 and 8 of the experiment and tissue specimens were prepared. mRNA was isolated from the samples, and its relative level was evaluated by electrophoresis. Complementary DNA was obtained by reverse transcriptase using MMLV reagent. Expression of the studied genes was evaluated according to the results of PCR real-time. The level of Ki-67 gene expression was normalized in relation to β-ACTIN gene. On day 4 there was a higher level of Ki-67 gene expression in the first group and on day 8 that was in the 2nd group. The analysis of the expression of Ki-67 gene as compared to β-ACTIN allows concluding that the drug Oxytocin improves corneal regeneration.
